Now that all 5 classes have been revealed, i thought i'd make a post to sum up and show some pictures.
Diablo3 Classes:

Skills:
Skills are spells or abilities that you use. You unlock new skills as you level up, as well as getting new points you can spend on these skills. Skills have caps, that can be increased throughout the game.
Traits:
Traits is a new system that will be replacing passive skills. This mechanic was added to allow players to customize their character without having to waste skill points to do so.
Runes:
In D2, runes are stones we put into our gear instead of gems, they can form rune words, transforming the item into a unique. In D3 runes are skill runes that you can insert into your skills, transforming and changing spells and skills completely. There are 7 levels of each rune type, increasing the effect of the rune.
Examples
Which Doctor's Blow Dart skill:
Regular - Blows out a dart
Crimson - Adds fire damage
Golden - Steals mana
Obsidian - Slows target
Indigo - Adds multiple darts
Alabaster- Blows out a snake that stuns the target
Barbarian's Throw Weapon skill:
Regular - throws a weapon for basic damage
Crimson - Adds damage
Obsidian - Turns into a stunning hammer
Indigo - Adds ricochet
Alabaster - Confuses target
Golden - Throw a corpse instead of a weapon
Wizard's Hydra skill:
Regular - Summons a fire spitting hydra
Crimson - Changes to short range frost
Indigo - Creates a never missing lightning hydra
Alabaster - Changes to a AoE arcane type
Obsidian - Adds acid element
Golden - Giant hydra that creates fire walls
Witch Doctor's Plague of Toads skill:
Regular - Summons out poison toads
Crimson - Changes them to fire damage
Golden - Reduces mana
Alabaster - Blinds target
Indigo - Causes the toads to rain down
Obsidian - Giant toad that eats monsters
Gear:
In D2, the gear reset look, there's about 6-8 tires, cycled 3 times. In D3 the gear progressively improves look, and there are 18 tires of gear. You can also dye parts of the gear, some dyes are rare and looks extra cool.
Gems:

Talisman:
A new type of inventory for your charms in D3, in D2 you had them in your normal inventory.
Crafting:
In D3 there's 3 people who can craft things for you. The blacksmith, mystic, and jeweler. You can get new recipes for them. They can even level up, and getting new looks for their caravans as they do. You can salvage gear, and deliver the materials along with some gold to get things crafted.
Health:
In addition to potions, you now get health blobs from mobs, keeping the phase up, not needing a trip to the town for every pack of mobs. There are new restrictions as well, you can't set up town portals when you want, only when you're out of combat.
Story telling:
In D2 you sometimes had to stand and wait, listening to the story being told to you, for over minutes at a time. In D3 you can listen to the story as you go, either by having a book read to you, or some NPC's talking to you as you slay
PvP:
You can enter a seperate place for PvP, Battle Arenas.
